景衣安神散穴位贴敷 对失眠大鼠行为学及相关细胞因子的影响∗ 

胡金霞1杨 帆2卢 军1王洪霞1江 钰1刘 涛1胡晓灵11 新疆维吾尔自治区中医药研究院,新疆 乌鲁木齐 830000;2 新疆军区总医院健康管理科 [摘 要] 目的:探讨景衣安神散穴位贴敷对失眠大鼠行为学以及细胞因子的影响。方法:将雄性大鼠 64 只按随机数字表法分为对照组、实验组、模型组、空白组,每组 16 只。对照组腹腔注射 0.9% 氯化钠溶液 1 mL/100 g),每日1次,连续注射2天,饲养7天。实验组、模型组、空白组均腹腔注射对苯丙氨酸(PCPA),每 日1次,连续注射2天制造睡眠剥夺模型。造模后模型组不进行任何干预,连续饲养7天。空白组将空白敷膜 贴于神阙、内关穴,每天1次,每次30 min,连续7天。实验组将景衣安神散敷贴于神阙、内关穴,每天1次,每 次30 min,连续7天。实验第8天采用酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)检测大 鼠血清白细胞介素1β(interleukin-1β,IL-1β)及肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)含量。 结果:实验组大鼠空旷实验得分下降,中央格停留时间减少,粪便颗粒减少(P<0.01),修饰次数增多(P<0.01),大 鼠挣扎次数较模型组减少,较空白组增加(P<0.01),绝望时间较模型组缩短,较空白组延长(P<0.01),实 验组大鼠血清IL-1β及TNF-α逐渐升高,差异有统计学意义(P<0.01)。结论:景衣安神散穴位贴敷可以升高失 眠大鼠血清IL-1β,TNF-α含量,诱导睡眠,恢复正常的睡眠-觉醒周期。 [关键词] 失眠;行为学;细胞因子;穴位贴敷;景衣安神散;动物实验 [中图分类号] R282 [文献标识码] A [文章编号] 2096-9600(2021)11-0041-05 41Western Journal of Traditional Chinese Medicine,2021 Vol.34 No.11 中 国 科 技 核 心 期 刊 药 理 研 究 ·

Methods: All 64 male rats were divided into the control group, the experiment group, the model group and the blank group according to random number table, 16 rats in each group. The control group accepted intraperitoneal injection of 0.9% sodium chloride solution (1 mL/100 g), once each day, for two consecutive days, feeding for seven days. The experiment group, the model group and the blank group received intraperitoneal injection of PCPA, once each day, for two consecutive days to set up sleep-deprivation models. After modelling, no intervention was given to the model group, and they were raised for seven consecutive days. The blank group applied blank film to Shenque (RN8) and Neiguan (PC6), once each day, 30 minutes each time, for seven days consecutively. The experiment group applied Jingyi Anshen powder to Shenque (RN8) and Neiguan (PC6), once each day, 30 minutes each time, for seven days consecutively. On the 8th day of the experiment, ELISA method was used to detect the contents of IL-1β and TNF-α of the rats. Results: The scores of open experiment decreased, the residence time of the central cell reduced, and fecal particles cut in the experiment group (P<0.01), grooming times increased (P<0.01), the number of struggles in rats reduced than the model group, in creased than the blank group (P<0.01), desperate time shortened than the model group, prolonged than the blank group (P<0.01), the levels of IL-1β and TNF-α rose in the experiment group, and the difference had statistical meaning (P<0.01). Conclusion: Jingyi Anshen powder acupoint application could lift the contents of IL-1β and TNF-α in the rats with insomnia, induce sleep, and return to normal sleep wake cycle.
